The first-ever clinical trial of sildenafil citrate, a phosphodiesterase type 5 inhibitor, in erectile dysfunction (ED) patients is set for Tuesday’s meeting, after the first clinical trial is announced by the Canadian Institute for Health Information and Research and the University of Toronto’s Health Sciences Centre.
The trial is the first major trial to compare the effects of sildenafil, an oral medication used to treat erectile dysfunction, with the effects of a drug that belongs to the same class of medications. “This is important because some of the side effects may be related to a lower blood pressure,” says Dr. David Chiu, associate professor of urology at Toronto's St. George’s Hospital, and the lead investigator of the study, Dr. Chiu. “Some of the side effects of this drug may be related to the changes in the vasculature.”
The researchers recruited 40 patients with ED who were taking sildenafil citrate (Viagra), a phosphodiesterase type 5 inhibitor, for at least three months. The researchers studied two main groups of patients: patients who were treated with sildenafil citrate and those who were not.
After the first clinical trial, the trial was stopped early because of the low response rate of the placebo group (less than 0.3%). The study is expected to last up to 5 years.
The researchers’ goal was to determine whether the clinical effects of sildenafil citrate, a phosphodiesterase type 5 inhibitor, were similar in the groups that received the drug.
“Sildenafil citrate was well tolerated with few adverse reactions in the group that received sildenafil citrate, and there were no major adverse events in the placebo group,” says Dr. “The main side effect profile of sildenafil citrate is similar to sildenafil citrate’s.
“In addition to the adverse reactions, there are also fewer severe adverse events in the placebo group, which is most likely due to the lower dose of the drug.”
